Unravelling the Therapeutic Potential of Antibiotics in Hypoxia in a Breast Cancer MCF-7 Cell Line Model

چکیده

Antibiotics inhibit breast cancer stem cells (CSCs) by suppressing mitochondrial biogenesis. However, the effectiveness of antibiotics in clinical settings is inconsistent. This inconsistency raises question whether tumor microenvironment, particularly hypoxia, plays a role response to antibiotics. Therefore, goal this study was evaluate five commonly used for inhibiting CSCs under hypoxia using an MCF-7 cell line model. We assessed number through mammosphere formation assay and aldehyde dehydrogenase (ALDH)-bright count. Additionally, we examined impact on stress membrane potential. Furthermore, analyzed levels proteins associated with therapeutic resistance. There no significant difference between cultured normoxic hypoxic conditions. did affect rate CSC inhibition Specifically, azithromycin unable sphere hypoxia. Erythromycin doxycycline not reduce ratio ALDH-bright cells, despite decreasing mammospheres. treatment chloramphenicol, doxycycline, tetracycline led overexpression resistance protein. Our findings suggest that may weaken inhibitory effects

The Activity of Sirtuin 1 in MCF-7 Breast Cancer Cell Line: The Effects of Visfatin

Background & Objectives: Breast cancer is the most common cancer and the second leading cause of cancer deaths among women. Obesity, hormones, and growth factors are the risk factors for this kind of cancer. One of the changes observed in patients suffering from breast cancer is the elevated Visfatin or nicotinamide phosphoribosyl transferase (NAMPT) in their tumor tissues and blood. The increa...

Cytotoxicity Effect of Cladribine on the MCF-7 Human Breast Cancer Cell Line

Cladribine, an analogue of deoxyadenosine, is highly toxic for both non-dividing and proliferating cells and has shown activity in the treatment of several malignancies. Therefore, the aim of the present study is to investigate the cytotoxicity effect of cladribine (2-CdA) on the breast cancer cell line, MCF-7 (estrogen receptor positive, ER+). MTT assay, annexin V-Fluorescein/PI and Hoechst 33...

CD44 expression changes and increased apoptosis in MCF-7 cell line of breast cancer in simulated microgravity condition

Introduction: Studies have shown that simulated microgravity (SMG) affects tumor cell proliferation and metastasis. However, the underlying mechanism and its molecular basis are still not well known. In recent years, due to the role of CD44 in breast cancer and its high expression in invasive basal tumors, it has been the subject of extensive research. There is a conflicting data on the role of...
